In this tutorial we look at how to interpret a NOESY NMR spectrum using a tripeptide as an example. 2D homogeneous NOE spectroscopy (NOESY) is a widely used experiment for looking at dipolar coupling between homonuclei such as H-H coupling. Because it relies on intramolecular (through space) interactions rather than through bond interactions it is extremely useful for assigning connectivities between spin systems and also for confirming the 3D structure of molecules.
